abstract = {Aquaporins are water channel proteins that facilitate water transport through the transcellular pathway, a route with high resistance due to lipid bilayer membranes. Aquaporins occur as multiple isoforms and plasma membrane aquaporins (PIPs) are known to modulate plant responses to abiotic stress such as drought. Previous studies have reported changes in PIPs expression in roots of Citrus plants under abiotic stresses; however, such changes are limited to grafted citrus trees with plant responses to environmental stresses being driven by two species. Therefore, the ability of a given Citrus species—i.e., non-grafted Citrus saplings—in modulating aquaporin expression under water deficit remains unknown. This study aimed to evaluate how water deficit changes aquaporin expression (PIP1; PIP2.1; and PIP2.5) and leaf gas exchange of saplings of three Citrus species: ‘Mandarin’ lime, ‘Swingle’ citrumelo and ‘Valencia’ sweet orange. Root aquaporin expression did not vary under water deficit or among species, but the predawn leaf water potential of ‘Valencia’ sweet orange was reduced due to water deficit. Decreases in stomatal conductance were found in all species under water deficit, which were associated with reduced PIP2.1 expression in leaves. However, ‘Mandarin’ lime exhibited the largest reduction in stomatal conductance and significant gene repression of not only PIP2.1 but also PIP2.5 in leaves under water deficit. ‘Mandarin’ lime is known as a drought tolerant rootstock widely used in citriculture and our data suggest that this rootstock presents a fast-response mechanism against drought based on stomatal closure associated with downregulation of PIP2.1 and PIP2.5 in leaves.},

abstract = {Boron (B) affects plasma membrane (PM) integrity and consequently modulates the P-type PM H+-ATPase activity creating a driving force for nutrient influx at the root level. Because citrus rootstocks respond differently to B supply, we hypothesised that PM H+-ATPase activity of varieties contrasting in horticultural traits would affect nutrient uptake by trees. Sweet orange (Citrus sinensis) trees grafted onto Rangpur lime (RL; Citrus limonia) or Swingle citrumelo (SW; Citrus paradisi × Poncirus trifoliata) were grown in nutrition solution with four B concentrations (0 [control concentration], 46, 230 and 460 μM B, as H3BO3) up to 7 days of treatment imposition after plant adaptation into the hydroponic condition. SW exhibited higher B absorption, leaf B and enzyme activity than RL. The highest enzyme activity was achieved with 230 μM of B 1 day after treatment imposition (ATI), whereas B excess impaired the PM H+-ATPase in all periods evaluated. Absorption of mineral nutrients correlated with PM H+-ATPase activity, with greater nutrient uptake per root unit in SW compared to RL. Leaf and root nutrient concentrations were equivalent to amounts absorbed and enzyme activity, with greater increments exhibited by trees grafted onto SW compared to RL. Effects of B supply on PM H+-ATPase activity explain distinct nutrient uptake patterns by trees, what supports fine-tuning fertilisation guidelines of citrus taking into account rootstock varieties.},

abstract = {Water scarcity can elicit drastic changes in plant metabolic and hormonal regulation, which may be of fundamental importance to stress tolerance. The study of plant the metabolic alterations in response to water deficit, especially the effects of the rootstocks level, is important to elucidate the mechanisms associated to drought tolerance. To verify the influence of rootstock and grafting on the tolerance to drought in citrus plants, we analyzed the growth, phytohormone levels and flavonoid profiles in grafted and ungrafted citrus plants subjected to different soil water regimes on plant status (well-watered, moderate drought and severe drought and rehydrated) under field conditions. The experiments were conducted under field conditions in the Brazilian Agricultural Research Corporation (EMBRAPA), Cruz das Almas, BA, Brazil. Water deficit reduced the total leaf area per plant in all canopy/rootstock combinations. Self-grafting reduce root volume, area and length when compared to ungrafted plants. Drought-induced increases in salicylic acid and abscisic acid associated with concomitant reductions in indoleacetic acid were observed in most canopy/rootstock combinations. However, plants with 'Sunki Maravilha' rootstocks exhibited the most pronounced changes in hormonal levels upon drought stress. Associated to these hormonal changes, drought also significantly affected flavonoid content and profile in both leaves and roots of the distinct citrus combinations. Glycosylated (GFs) and polimethoxylated flavonoids were predominantly found in leaves, whereas prenylated coumarins were found in the roots. Leaf levels of GFs (vicenin, F11, rutin and rhoifolin) were particularly modulated by drought in plants with ‘Rangpur Santa Cruz’ lime rootstock, whereas root levels of prenylated coumarins were most regulated by drought in plants with the 'Sunki Maravilha' root system. Taken together, these data indicate that the impacts of water deficit restriction on growth, hormonal balance and flavonoid profiles significantly varies depending on the canopy/rootstock combinations.},
