abstract = {The Asian citrus psyllid, Diaphorina citri Kuwayama, transmits the bacteria Candidatus Liberibacter associated with huanglongbing (HLB), a devastating disease of the citrus industry. The use of genetically modified plants is an alternative to control this vector. Conversely, technology based on RNA interference (RNAi) for silencing specific genes of a target insect could be attempted. This work evaluated the knockdown effect of the target genes calreticulin (DcCRT), laccase (DcLAC), and Snf7 (DcSnf7) by RNAi through feeding D. citri in Murraya paniculata leaves after the uptake of an aqueous solution with dsRNA homologous to each vector target gene. Confocal microscopy revealed the uptake of the fluorescent-labeled dsRNA by detached leaves and the symplastic movement, allowing the ingestion by the feeding insect. A reduction in the survival rate was observed only 144 h after the beginning of feeding with dsRNA targeting DcSnf7; however, no reduction in transcript accumulation. The knockdown of the DcCRT and DcLAC genes was detected only 12 and 96 h after insect feeding, respectively. Additionally, a reduction in amino acid excretion from insects fed with dsRNA targets to DcCRT and DcLAC was observed 120 h after the beginning of feeding. However, the effects of the dsRNAs tested here appear to be minimal, both at the transcriptional and phenotype levels. For most concentrations and time points, no effects were observed. Therefore, the knockdown of genes DcCRT, DcLAC, and DcSnf7 do not appear to have the potential to control of D. citri through RNAi-mediated gene silencing.},

We evaluated the effects of micronutrient (Zn, Mn, and/or Cu) supply and ‘Candidatus Liberibacter asiaticus’ (CLas) infection on physiological and growth traits of sweet orange trees (Citrus sinensis (L.) Osbeck)) to understand if enhanced micronutrient supply would improve the growth of plants infected with CLas and reduce bacterial infection, as well as the acquisition of CLas by adults and nymphs of Diaphorina citri, correlated with nutritional status of trees. Plants were either grafted with buds obtained from micrografted plants (healthy, −) or buds infected with CLas (diseased plants, +). Infected plants were exposed to one of the five nutrient treatments, applied to leaf and root: (i) nilZnMnCu+: not fertilized with Cu, Mn, and Zn; (ii) Zn+: fertilized with zinc sulfate (ZnSO4.H2O); (iii) Mn+: fertilized with manganese sulfate (MnSO4.H2O); (iv) Cu+: fertilized with copper hydroxide [Cu(OH)2]; and (v) ZnMnCu+: fertilized with all three micronutrients. Likewise, healthy plants were exposed to one of the two treatments as above: (i) nilZnMnCu− or (v) ZnMnCu−. We found that CLas impairs plant biomass production regardless of nutrient treatments, especially root growth, and increases specific leaf dry weight as disease progresses because of starch accumulation. Moreover, individual supply with Zn, Mn, or Cu can mitigate such deleterious effects of HLB on starch metabolism. HLB also changes nutrient concentrations in both leaves and sap extract, regardless of nutrient treatments, although treatments do not reduce the CLas titer in plants as determined by RT-qPCR. On the contrary, micronutrients applied in combination (ZnMnCu+) can reduce the acquisition of CLas by adults and especially nymphs of D. citri, likely reducing the disease infection in citrus orchards.},

abstract = {Bacillus thuringiensis (Bt) toxins are effective in controlling insect pests either through the spraying of products or when expressed in transgenic crops. The discovery of endophytic Bt strains opened new perspectives for studies aimed at the control of sap-sucking insects, such as the Asian citrus psyllid Diaphorina citri Kuwayama (Hemiptera: Liviidae), a vector of “Candidatus Liberibacter spp.,” associated with citrus huanglongbing (HLB). In this study, translocation of endophytic Bt strains in citrus seedlings inoculated with Bt suspension delivered by soil-drench, and their systemic pathogenicity to D. citri nymphs were investigated. The pathogenicity of three wild-type Bt strains against D. citri third-instar nymphs was demonstrated. Among the 10 recombinant strains tested (each of them harboring a single cry or cyt gene), 3 can be highlighted, causing 42%–77% and 66%–90% nymphal mortality at 2 and 5 d after inoculation, respectively. The isolation of Bt cells from young citrus shoots and dead nymphs, and PCR performed with specific primers, confirmed the involvement of the Bt strains in the psyllid mortality. This is the first report showing the translocation of Bt strains from citrus seedling roots to shoots and their potential to control D. citri nymphs that fed on these soil-drench inoculated seedlings. The Bt strains that caused the highest mortality rates have the potential to be used as bioinsecticides to control D. citri and the identified genes can be used for the production of transgenic Bt citrus.},

abstract = {Interactions between insects and plant pathogens have been more enthusiastically studied in the recent decade, especially those relationships which takes the insects as vectors. The spectrum of these interactions ranges from mutualistic to pathogenic. The length of the co-evolutionary process will determine whether a microorganism shares a friend or a foe relationship with its host, and a friendship connection is frequently observed if the coexistence is longer. This review updates knowledge about the morphological, physiological and genetic mechanisms that drive the interaction between ‘Candidatus Liberibacter asiaticus’ (Las) and its vector, the Asian citrus psyllid, Diaphorina citri. Las is the predominant causal agent of citrus huanglongbing (HLB) disease, the major constrain to citrus production worldwide. This bacterium is transmitted by D. citri, in a propagative-circulative manner during its feeding from plant host. Understanding of the interactions among vector, plant pathogen and host plant are important for the management of this vector-borne disease complex.},

abstract = {Huanglongbing (HLB) is a severe disease of citrus caused by the bacterium Candidatus Liberibacter. In America, the most common species is Candidatus Liberibacter asiaticus (CLas). In a previous study of the Citrus-HLB pathosystem, our group found differences in CLas multiplication in a population of hybrids obtained by hybridization between Citrus sunki Hort. Ex Tan. and Poncirus trifoliata (L.) Raf. cv. Rubidoux. The bacterial concentration was higher in C. sunki than in P. trifoliata. Thus, this study aims to map phenotypic (QTL) and gene expression (eQTL) data associated with host response to CLas in the linkage groups (LGs) of the previously constructed parental maps of C. sunki and P. trifoliata cv. Rubidoux. For the phenotypic analysis, we used a population of 79 F1 hybrids (termed citrandarins) between C. sunki and P. trifoliata. CLas and starch were quantified in the leaves of the plants two years after pathogen inoculation, allowing the classification of hybrids as resistant, tolerant, and susceptible. The expression of 14 candidate genes was measured in 72 hybrids of the population and used as expression data for the eQTL mapping. We located nine QTL and 52 eQTL in the C. sunki map and 17 QTL and 40 eQTL in the P. trifoliata map. The overlap of eQTL of the majority of genes with QTL from the phenotypic data indicates that the genes are related to the phenotype and are probably related to pathogen infection.},

abstract = {Environmental factors that influence flight activity of Diaphorina citri Kuwayama (Hemiptera: Liviidae) may have implications for Huanglongbing spread and management. In this work, four studies were conducted to evaluate the effect of environmental conditions on D. citri take-off. In the first, insects were transferred to sweet orange seedlings and confined inside an acrylic cage to verify the take-off periodicity and the effect of environmental factors on this process. In the second, take-off temperature threshold was estimated by recording the number of insects that initiated flight from a platform when subjected to gradual temperature increases from 15 to 39°C. In the other studies, we evaluated the effect of different photoperiods and temperature regimes (third study) and of constant temperatures (fourth study) on the propensity for D. citri flight. Insects were confined in clear plastic bottle cages with tubes of 50 ml placed on the cab, to collect emerged adults that initiated flights. Results showed that a small portion of the tested population (maximum 10%) tends to take off from plants and this behaviour is more prevalent in the afternoon (14:00–16:00 h), coinciding with daytimes of lower humidity and higher thermal amplitude. Adults that were submitted to lower temperatures (18°C) and short light periods (10 h) showed less propensity to flight. In contrast, at constant 27°C, the insects were more prone to flight, and this result was confirmed when individuals were submitted to increases in temperature, indicating that 27.14°C is the take-off temperature threshold of D. citri. Results show that temperature plays an important role in the flight activity of D. citri and suggest that control measures of the insect may be more effective in the morning and in temperatures below 27°C, when the probability to take off from a host is lower.},

abstract = {ABSTRACT The use of promoters preferentially expressed in specific plant tissues is a desirable strategy to search for resistance for pathogens that colonize these tissues. The bacterium Candidatus Liberibacter asiaticus (Las), associated with huanglongbing disease (HLB) of citrus, colonizes phloem vessels. Some promoters, besides conferring tissue-specific expression, can also respond to the presence of the pathogen. The objective of the present study was to verify if the presence of Las could modulate the activation of the phloem-specific promoters AtPP2 (Arabidopsis thaliana phloem protein 2), AtSUC2 (A. thaliana sucrose transporter 2) and CsPP2 ( pCitrus phloemrotein 2), known to be expressed in Citrus sinensis phloem. ‘Hamlin’ sweet orange plants (Citrus sinensis L. Osbeck) transformed with the uidA (GUS) reporter gene under the control of AtPP2, AtSUC2 and CsPP2 promoters were infected to evaluate the interdependence between transgene expression and the concentration of Las. Plants were inoculated with Las by Diaphorina citri and eighteen months later, bacterial concentration and uidA expression were determined by qPCR and RT-qPCR, respectively. Reporter gene expression driven by AtSUC2 promoter was strongly and positively correlated with Las concentration. Therefore, this promoter combines desirable features of both tissue-specificity and pathogen-inducibility for the production of transgenic plants tolerant to Las.},
